Immune system is the body's defense against foreign organisms and harmful chemicals. Cyclosporine (CsA) is an immunosuppressant drug widely used in the treatment of many diseases. Although the therapeutic effect of CsA, it has many adverse effects. Echinacea purpurea roots extracts (EPR) is used as an immune-enhancing plant. The present study aimed to evaluate the effect of EPR against immunosuppressive induced by CsA. Thirty two male albino rats were divided into control, CsA (immunosuppressive model); rats were injected subcutaneously (Sc) with CsA at a dose level of (15 mg/kg) for 21 days, CsA + EPR (100 mg/kg/day orally) and CsA + EPR (200 mg/kg/day orally). At the end of the experiment (21 days) blood and tissues samples was collected. Complete blood count, serum levels of interleukin 1 alpha (IL-1α) and tumor necrosis factor (TNF)-α were meaused. As well as the enzymatic antioxidant activities of superoxide dismutase (SOD) and glutathione-s-transferase (GSTs) were estimated in splenic tissues. Histopathological and immunohistochemically examinations were done on the spleen sections. The CsA treatment induced a significant decrease in RBCs, WBCs, splenic antioxidant enzymes activities (SOD and GSTs), and serum inflammatory cytokines (TNF-α and IL-1α). There was a marked decrease in the cellularity with congested blood sinusoids, depletion of the brown stained CD3 positive T lymphocytes and a marked decrease in the CD56 positive Natural Killer Cells in examined the spleen sections. EPR significantly ameliorated the changes in hematological, biochemical histopathological and immunohistochemistry CD3 and CD56 results compared with CsA group. Therefore, EPR has an immunostimulant and antioxidant effects on the spleen that opposing the immunosuppressive effect of CsA.
