Abstract
:
The aim: To assess the effect of recombinant human bone morphogenetic protein-2 (BMP-2) with different concentration of poly lactide-co-glycolide (PLGA), ploycaprolacton (PCL) and PLGA type on bone regeneration in closed bony defects in rabbits using histology and Micro CT images. Materials and Methods: Sixty, 5 weeks old, New Zealand rabbits used in this study, divided randomly into 3 groups A, B and C according to the formula assigned formula A (PLGA 30%, PCL 15%, PLGA type 75:25), formula B (PLGA 10%, PCL 5%, PLGA type 75:25) and formula C (rhBMP-2 in normal saline). Result: Group A showed the highest amount of bone formation 85.16% and complete healing of the defect wall than group B 48.14% and C 19.47%. These results were confirmed by micro-CT findings in which group A showed the highest new bone formation and complete healing of the defect wall when compared to group B (partial healing of the defect wall and less bone formation) and group C ( incomplete healing of the defect wall and lowest new bone formation). Conclusion: Optimum bone formation was achieved using formula A which had lower initial burst and prolonged drug sustained in the defect site 78%.
